چکیده

Drug idiosyncrasy is an adverse event of unknown etiology that occurs in a small fraction of people taking a drug. The histamine2 (H2)-receptor antagonist ranitidine causes idiosyncratic reactions in patients. To investigate the hypothesis that ranitidine could induce hematological toxic effects, the drug was administered intraperitoneally (ip) to two of six groups of 200-220 g male Wistar rats (n=6). Group I received as single dose of saline solution (NaCl, 200 μL) Group II received 200 μL of NaCl, ip, at 0, 24, 48 and 72 h, Group III (controls of the vehicle) received as single dose of dimethyl sulfoxide (DMSO, 200μL), ip, Group IV (controls of the vehicle) received 200 μL of DMSO, ip, at 0, 24, 48, and 72 h, Group V received a single dose of 100 mg/kg of ranitidine in 200 μL of DMSO) ip, Group VI received 50 mg/kg of ranitidine, in 200 μL of DMSO, ip, at 0, 24, 48, and 72 h. Erythrograms, leucograms were measured; percent methemoglobin content of the blood was analyzed spectophotometrically. Methemoglobinemia increased to a significant extent following ranitidine treatment, in both study groups. White blood cell and neutrophil counts showed a discrete reduction following either regime of treatment. A marginal mobilization of the neutrophil pool probably occured. Following the multiple dose regime, leukocyte counts increased, but their distribution maintenance profile remained the same. The administration of ranitidine to rats induces methemoglobinemia in the administered doses, suggesting that from a certain concentration and dosage scheme the substance can be methemeglobinemia inductor. *Correspondence: W. The effect of Matricaria Chamomilla on the treatment of ibuprofen-induced gastric ulcers in male rats

ABSTRACT Introduction: Formation of gastric ulcer is a multifactorial process so that vascular changes, mucous secretion, gastric acid and gastrointestinal tract mucous biochemistry are involved in gastric ulcers. Despite therapeutic properties, ibuprofen as a non-steroidal anti-inflammatory drug (NSAID) can cause mucosal lesions in the stomach. In this study, the effect of Matri...

Bioprotective Efficacy of Erucin Against 7,12-Dimethylbenz(α)anthracene-Induced Microstructural Changes in Male Wistar Rats.

OBJECTIVE Environmental pollutants are responsible for inducing a number of biochemical and physical changes in the various organs of an organism. The most potent pollutants is7,12-dimethylbenz(α)anthracene (DMBA) (a genotoxic carcinogen), a polyaromatic hydrocarbon. It is known to induce a number of genetic changes including the formation of DNA adducts. These genotypic changes in turn are res...

Comparison of the metabolism and toxicity of dapsone in rat, mouse and man.

The metabolism and toxicity of dapsone was compared in vitro and in vivo in rat, mouse and man. Metabolism was assessed by high-pressure liquid chromatography-mass spectrometry and methemoglobin formation has been used as a toxic endpoint. The greatest toxicity in vitro was seen in microsomes prepared from male Wistar rats (36.6 +/- 1.5% methemoglobin), although toxicity was also seen in micros...

The Effect of Scopolamine on Avoidance Memory and Hippocampal Neurons in Male Wistar Rats

Introduction: Cholinergic systems are involved in learning and memory. Scopolamine, a muscarinic acetylcholine receptor antagonist, is used as a standard/ reference drug for inducing cognitive deficits in healthy humans and animals. The purpose of this study was to evaluate the effects of scopolamine on avoidance memory and number of neurons in rat’s hippocampus. . Methods: Thirty five male alb...

Effect of Pithecellobium dulce (Roxb.) Benth. fruit extract on cysteamine induced duodenal ulcer in rats.

The edible fruits of Pithecellobium dulce (Roxb.) Benth. are traditionally used for various gastric complications in India. Here, we investigated the antiulcer activity of hydroalcoholic fruit extract of P. dulce (HAEPD) by applying cysteamine induced duodenal ulcer model in rats. Duodenal ulcer was induced in male albino Wistar rats by oral administration of cysteamine @ 420 mg/kg body wt. as ...
